You will love it.
The wife (I actually proposed in Barcelona at Placa Reail) spent two weeks there last August - one week in Barcelona one week in Madrid.
As others have said, Barcelona is beautiful. And the people match it. Definitely hit up the Gaudi spots - Parc Guell, Casa Mila and definitely the Sagrada Familia. Word to the wise on the Sagrada - eat before you go. Lines tend to be long and I found there wasn't much in terms of places to eat other than fast food and very touristy trap places.
Las Ramblas is cool - but I wouldn't spend a ton of time on it. Beware, as pickpocketers in this area are the worst. Keep your valuables in your front pocket and you will be fine.
Spend a ton of time in the Barri Gotic (Gothic Quarter). We literally spent two days just wandering around there and ducking into the bars and little restaurants within all the side alleys. Two great places we found: Milk (owned by a nice Irish lady, great specialty cocktails and good comfort food) and Sensi Bistro (awesome tapas).
The beach is beautiful. As others have said, bathing topless (and I saw a few fully nude folks) is common.
If you or your wife are shoppers, Barcelona is a dream. Tons of H&Ms and Zara. Zara is great. Wish they had more in the States.
If you get a chance, Google classic Spanish guitar and try to see a show. We booked an evening show at a church in the Barri Gotic and it was really great. They usually serve wine as well during the show.
Beers - in Barcelona, you will likely drink a lot of Estrella. Very good. You may find Mahou (my personal favorite), but they its more prominent in Madrid. San Miguel is acceptable. Cruzcampo is also good.
Transport -Barcelona is very walkable actually for the most part, but it will wear you out. As others have said, the Metro is cheap and easy to learn to ride.
I unfortunately didn't make it to the Olympic village or the mountains, which we'll make sure to hit on our next trip.
Anyway, I could go on and on.

Place is great. As mentioned before, see as much Gaudi as you can (There are like 18 different Gaudi places). See The Barcelona Cathedral (iirc they have a kindof show in front of it at about noon) and the Sangrada Familia. Grab the Rick Steve's book, it has a bunch of walking tours that you can do to make sure you see a most of the stuff in each of the areas.
Try to catch a flamenco show (there are a ton around La Ramblas. It is usually male and female dancers to classical guitarists and a singer wailing in Catalunian. We went to Seville to see what was supposed to be the best flamenco dancing in Spain, but I enjoyed it more in Barcelona.
And the Bari Gotic was a really awesome part of town. It also has the Pablo Picasso museum which is worth it, if museums are your thing.

Museums are world class. Rent a car and travel the Costa Del Sol. I am a huge fan of SOuthern Spain, much more laid back and the people are truly friendly. Try the garlic chicken!

I was there more years ago than I care to count. It is beautiful, the food is delicious and the Spanish women are by far the best looking in Europe.
