Chef's Eulogy
This was probably the most serious moment in South Park history. As ridiculous as the episode was the eulogy given by Kyle really showed that the creators cared for IH and wished things had turned out better then they did.

I also think there was another when the congressman who Colbert got to say he liked cocaine got a lot of flack, Colbert talked good about him on air in a serious moment.

a lot of the shows mentioned in this thread either definitely had serious moments or are not sitcoms. the only 30 minute non-cartoon shows i can think of that have never had a serious moment are:
seinfeld
arrested development
always sunny in philadelphia

1. Al and the family lose the Dodge at the drive through car-wash because it was originally red(the dirt washed off). Al was worried throughout the entire episode because his beloved car was gone, but to everybody's surprise, he was afraid that his family photo, which was stuffed inside a copy of Big-un's, was lost. That was serious.
2. Al was mad a Peggy, so he spent the evening at his friend from the shoe store (white playboy dude). Al was beging tempted by a hot airline attendent, but he turned her down because he actually loved his wife. That was serious.

The band breaks up numerous times, and Jemaine always gets dumped by his girlfriends.
