Just for reference, he edited his post so that list seems more sane than what he originally had.
I mean do what you want. Everyone is different.
Flying overnight to London, possibly getting there mid day, and then leaving a day and a half later seems horrible to me, but everyone is different.

Again, the flights won’t be the problem. Fly for free. Just looking for recommendations of places to check out.

I get this response but if you are someone who operates fine with not a ton of sleep its not that big of a deal for a few days. If you can only get away for a few days it is what it is while not ideal it can still be great.
Jetlag kills my wife so these types of ideas always get shot down for me.

That's a game changer, especially these days.
I like to hit the ground running in places and can go on little sleep. I think London would be a good choice. I had a similar solo trip there years ago as part of a work trip and really enjoyed myself. Got to see a few major sites and sit and enjoy people watching at a couple pubs.

But why? The flying time vs vacation time ratio is way off. I don’t see how that would be enjoyable.
Maybe Caribbean, Central America, Canada makes more sense. You could probably do Machu Picchu if that was all you wanted to do in Peru. Or a badass scuba trip in Belize. Or hiking in Banff.

Look, I don't dispute that this can work (I landed in Sydney, Australia at 10AM on a Tuesday, went to the proposal on Wednesday, and flew out Friday morning,) but unless your wife is ex military, that's a tough slog with jet lag.
Most of the flights out of IAH to Europe seem to be early afternoon flights, so a connection wouldn't be difficult from MSY or similar.
Problem is, direct flights from IAH (for instance) to Manchester are 9 hour flights (direct on Singapore.) Ireland is a possibility too, but examine flying into SNN instead of DUB. Other than the Post Office and the St. James Gate brewery, Dublin is much less impressive than the stuff I found in the west right outside of SNN. You can get to SNN on Delta via ATL, too.
I guess the question is, what is it you're intending to do once you get there? I'm a big fan of impromptu, "just because" trips overseas, but you could waste all of day 3 in the British Museum or shopping in Picadilly, and for what? Not to mention, landing in LHR late morning means you're getting to the hotel mid afternoon (clearing immigration and transit), which essentially just leaves you time to eat dinner.
If you're focused on London, given your username, check out a bar in SoHo called "Crobar." It's a metal bar that I was at for the "frick you, I'm Millwall" terrorist attack nearby.

Do a domestic trip as direct a flight as you can.
eta: after reading the rest of the OP I still don't understand a 9+ hour flight (or connecting flights) for such a short trip. I would do short flights to cities you have already been to, rent a car, get an airbnb or hotel in a "close" driving distance and not spend so much time traveling.

i can leave lafayette at 7am, and be in cozumel at the hotel at 2pm. Snorkel all afternoon. Then 2 days of diving all day. The flight home on the last day is at 2pm so you can snorkel or hang out at the pool till 11am.

Heck, I did this as a daytrip two years ago. I got on an early flight from MSY, got my rental car at DEN around 9:00 AM, drove Trail Ridge Road, and got back to DEN about 7:30 PM for my flight back to MSY.
It was an awesome day, and it's fun to travel without luggage. I've done similar daytrips to Chicago on eight occasions; the flights can be cheaper than the hotel, and sometimes, it's fun to have a one-day vacation.
METAL, I like your premise and your style. Ignore the haters. I often like to travel quickly. Sometimes, a short trip can help me decide whether or not I'd like to make a longer one. I'll post some recs.

I think you could make roatan or curacao work. Caribbean islands make the most sense to me as you wont have much ground to cover when you get there.

- In Canada, the person who said Quebec City is absolutely correct. I was there in winter, and that was fun (riding a dogsled to the Ice Hotel, etc.), but I would love to go back in summer.
I've never been to the Canadian Rockies, but that would be my second destination. You could fly to either Calgary or Edmonton and drive west for just a bit, either in summer or in winter.
- In Mexico, let me start by saying that Mexico City is awesome. The cathedral and Zocalo area is worth a day.
Next, the national anthropology museum is one of the world's best museums, but it's really big, so pay $60 or so for a guide to take you through the most spectacular areas. Take your wife to the Ballet Folklorico -- the theater itself is worth the price of admission, and you'll hear some great mariachi. Go to Chapultepec and see Maximillian's palace. Finally, get out of the city and go to Teotihuacan to see the massive temples. If you've got more time, there's lots of Frida stuff, Leon Trotsky's house, the holocaust museum, and Xochimilco.
So, Mexico City is 4 or 5 days at least, but you can split it up, since it's so close.
I love Puebla, and three days is perfect for it.
I've never been to Oaxaca, but it would be my next choice.
- In Belize, three days is perfect for either a land trip or an island trip.
On land, you simply must see the ATM Cave. You can drive out to a monkey preserve, check out Mayan ruins at Altun Ha, etc. with another day, drive into Guatemala and see the greatest Mayan ruins at Tikal (temples at the end of Star Wars movie)
- Guatemala is perfect for a 3-day trip. The amazing colonial city of Antigua is only 45 minutes from the airport, and from there you can go on to Lake Atitlan. You can "see" each of those places in a day.
- Panama City is a perfect place for a long weekend. The canal is cool, the Casco Antiguo area is charming and old, and Sobernia National Park is the best rainforest that I've seen.
- In the Caribbean, I want to recommend Barbados. It's got a unique culture and great people. Lots of the Caribbean is just resorts and mass tourism and becoming homogenous, but that's not the case in Barbados. Great snorkeling and diving too.

Europe seems like a stretch for a very short trip. How about Toronto? I’ve never been but heard it’s supposed to be a cool city.
