I'm not going to qualify it as the greatest ever, but the drug deal scene of boogie nights is excellent. The actors appear so nervous and it is amplified by the Chinese kid lighting firecrackers inside. The guy is so blown over on freebase coke, and is hilarious when he tries to sing "sister Christian" and "jesse's girl." Russian roulette. John C Reilly's hilarious reaction to drug dealers carrying guns and walberg's awesome acting while staring.

Almost listed this one... It's up there... Way up there... It's incredible... Roger Ebert even said it some of the best scenes ever filmed in his At The Movies review in 1997. Siskel and Ebert - Boogie Nights
Fun fact:
P.T. Anderson kept Wahlberg awake for like 2 or 3 straight days before shooting that scene so he would look so strung out... That shot where the camera pans in and Wahlberg totally zones out was not scripted.. He was genuinely "out of it" for that 10 seconds or so... He was mentally trapped out

I know this pic is art and not the actual movie scene, but for me... I sat in the theater in this scene with goosebumps thinking this is why movies exist. For scenes like this...

Final scene of Last of the Mohicans. Can't believe it took 4 pages.
Love the ending of the Prestige by the way. Just throwing that in for shits and giggles.

Are we talking greatest from a filmmaking/storytelling aspect, or the most famous/memorable scene? Probably a huge difference between the 2.
If most memorable, it's probably something out of wizard of oz, gone with the wind or vader "I am your father"

The chase sequence through the sewers in The Third Man. Harry Lime's fingers reaching through the grate... perfection.
Kim Novak emerging from the bathroom, dressed as Carlotta Valdez again, bathed in a mysterious glow in Vertigo.
The steadicam sequence in Children of Men.
The final scene of Casablanca.
The steely tension of the cafe sequence in Inglorious Basterds.
