No lie, but my neighbor did the ancestry DNA last month and got an email from some guy in LA who also shares her exact profile other than the Y chromosome obviously. Come to find out, my neighbors father is actually her fricking grandfather

These are common misconceptions. Ultimately, the key is registration with a federally recognized tribe. And many of those tribal councils are reticent to accept new members because a lot of folks are interested getting a cut of the casino cash. But if the OP's uncle has papers that his ancestor had, that would help his case with whatever tribe she belonged to.

Apparently, the Cherokee tribe require you have evidence of lineal descent from a known Cherokee regardless of blood quantum. If you have access to birth certificates from your great grandmother down to you that qualifies you for admittance to the Cherokee nation.

Application to the Cherokee nation.

BTW, if you great grandparent was 100% Cherokee, you would be 1/8, not 1/16

My mother's great grandmother, my great-great grandmother.
